The Chemokine Receptor CCR3 Is Potentially Involved in the Homing of Prostate Cancer Cells to Bone: Implication of Bone-Marrow Adipocytes

Bone metastasis remains the most frequent and deadliest complication of prostate cancer (PCa). Mechanisms leading to homing tumor cells bone remain poorly characterized. Role chemokines in providing navigational cues migrating bearing specific receptors is well established. an adipocyte-rich organ since 50 70% adult marrow (BM) volume comprise adipocytes (BM-Ads), which are likely produce within microenvironment. Using vitro migration assays, we demonstrated that soluble factors released by human primary BM-Ads able support directed PCa a CCR3-dependent manner. In addition, showed CCL7, chemokine previously involved outside gland, BM-Ads. These effects amplified obesity ageing, two clinical conditions known promote aggressive metastatic PCa. tumors, found enrichment CCR3 vs. tumors at mRNA levels using Oncomine microarray database. immunohistochemistry experiments overexpression versus visceral metastases. results underline potential importance process imply CCR3/CCL7 axis whose pharmacological interest needs be evaluated.
